Sigatech DP 50/5mg Tablet Product Description
Composition: Each film-coated tablet contains:
- Sitagliptin: 50 mg (as Sitagliptin Phosphate Monohydrate)
- Dapagliflozin: 5 mg
Therapeutic Class: Antidiabetic combination (DPP-4 inhibitor + SGLT2 inhibitor)
Indications: Sigatech DP 50/5mg Tablet is used in the treatment of type 2 diabetes mellitus in adults to improve glycemic control when diet and exercise alone are inadequate, or in combination with other antidiabetic medications (e.g., metformin, sulfonylureas, or insulin) when these do not provide sufficient blood sugar control. It helps reduce blood glucose levels and may lower the risk of cardiovascular events like heart attack or heart failure.
Mechanism of Action:
- Sitagliptin: A dipeptidyl peptidase-4 (DPP-4) inhibitor that increases insulin secretion by enhancing incretin levels (GLP-1 and GIP). It reduces glucagon production, thereby decreasing hepatic glucose output.
- Dapagliflozin: A sodium-glucose cotransporter 2 (SGLT2) inhibitor that blocks glucose reabsorption in the kidneys, promoting urinary glucose excretion and reducing blood glucose levels independently of insulin.
Dosage and Administration:
- Typically, one tablet is taken orally once daily, with or without food, as prescribed by a healthcare provider. Dosage may vary based on the patient’s condition and renal function. Consult a doctor for specific instructions.
- For patients with moderate renal impairment (GFR 30–45 ml/min), sitagliptin dosage may be adjusted to 50 mg, and dapagliflozin use should be evaluated. Not recommended for severe renal impairment (GFR <30 ml/min) without medical supervision.
Packaging: Available in strips of 10 tablets.
Side Effects:
- Common: Vaginal yeast infections, urinary tract infections (UTIs), nasopharyngitis, headache, nausea, diarrhea.
- Serious (rare): Hypoglycemia (especially when combined with other antidiabetic drugs), pancreatitis, diabetic ketoacidosis (DKA), dehydration, low blood pressure, acute kidney injury.
- Patients should monitor for symptoms like severe abdominal pain (pancreatitis), excessive thirst, frequent urination, or dizziness and seek immediate medical attention if these occur.
Precautions and Warnings:
- Contraindications: Hypersensitivity to sitagliptin, dapagliflozin, or any excipients; type 1 diabetes; diabetic ketoacidosis.
- Renal Impairment: Use with caution in patients with kidney issues; regular renal function monitoring is recommended.
- Pregnancy and Breastfeeding: Safety not established; consult a doctor.
- Hypoglycemia Risk: Increased when used with insulin or sulfonylureas; dose adjustments may be needed.
- Dehydration: Dapagliflozin may cause fluid loss; ensure adequate hydration.
